N-glycosylation of proteins is initiated in the endoplasmic reticulum (ER) by the transfer of the preassembled oligosaccharide glucose-3-mannose-9-N-acetylglucosamine-2 from dolichyl pyrophosphate to acceptor sites on the target protein by an oligosaccharyltransferase complex. This core oligosaccharide is sequentially processed by several ER glycosidases and by an endomannosidase, such as MANEA, in the Golgi. MANEA catalyzes the release of mono-, di-, and triglucosylmannose oligosaccharides by cleaving the alpha-1,2-mannosidic bond that links them to high-mannose glycans. The deduced 462-amino acid protein contains a putative type II transmembrane domain near the N terminus and shares about 77.9% identity with rat liver Manea.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate MANEA in samples. An antibody specific for MANEA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMANEA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MANEA is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MANEA bound in the initial step. The color development is stopped and the intensity of the color is measured.
