Intended Uses: This LCN2/NGAL ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human LCN2/NGAL. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: LCN2/NGAL ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-LCN2/NGAL antibody and an LCN2/NGAL-HRP conjugate. The assay sample and buffer are incubated together with LCN2/NGAL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the LCN2/NGAL concentration since LCN2/NGAL from samples and LCN2/NGAL-HRP conjugate compete for the anti-LCN2/NGAL antibody binding site. Since the number of sites is limited, as more sites are occupied by LCN2/NGAL from the sample, fewer sites are left to bind LCN2/NGAL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The LCN2/NGAL concentration in each sample is interpolated from this standard curve.

Uniprot Description

LCN2: Iron-trafficking protein involved in multiple processes such as apoptosis, innate immunity and renal development. Binds iron through association with 2,5-dihydroxybenzoic acid (2,5- DHBA), a siderophore that shares structural similarities with bacterial enterobactin, and delivers or removes iron from the cell, depending on the context. Iron-bound form (holo-24p3) is internalized following binding to the SLC22A17 (24p3R) receptor, leading to release of iron and subsequent increase of intracellular iron concentration. In contrast, association of the iron-free form (apo-24p3) with the SLC22A17 (24p3R) receptor is followed by association with an intracellular siderophore, iron chelation and iron transfer to the extracellular medium, thereby reducing intracellular iron concentration. Involved in apoptosis due to interleukin-3 (IL3) deprivation: iron-loaded form increases intracellular iron concentration without promoting apoptosis, while iron-free form decreases intracellular iron levels, inducing expression of the proapoptotic protein BCL2L11/BIM, resulting in apoptosis. Involved in innate immunity, possibly by sequestrating iron, leading to limit bacterial growth. Belongs to the calycin superfamily. Lipocalin family. 2 isoforms of the human protein are produced by alternative splicing.
