Principle of the Assay: L1CAM ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-L1CAM antibody and an L1CAM-HRP conjugate. The assay sample and buffer are incubated together with L1CAM-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the L1CAM concentration since L1CAM from samples and L1CAM-HRP conjugate compete for the anti-L1CAM antibody binding site. Since the number of sites is limited, as more sites are occupied by L1CAM from the sample, fewer sites are left to bind L1CAM-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The L1CAM concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is an axonal glycoprotein belonging to the immunoglobulin supergene family. The ectodomain, consisting of several immunoglobulin-like domains and fibronectin-like repeats (type III), is linked via a single transmembrane sequence to a conserved cytoplasmic domain. This cell adhesion molecule plays an important role in nervous system development, including neuronal migration and differentiation. Mutations in the gene cause X-linked neurological syndromes known as CRASH (corpus callosum hypoplasia, retardation, aphasia, spastic paraplegia and hydrocephalus). Alternative splicing of this gene results in multiple transcript variants, some of which include an alternate exon that is considered to be specific to neurons. [provided by RefSeq, May 2013]
