INTENDED USE This KLK7 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep KLK7. This ELISA kit for research use only!

Principle of the Assay: KLK7 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-KLK7 antibody and an KLK7-HRP conjugate. The assay sample and buffer are incubated together with KLK7-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the KLK7 concentration since KLK7 from samples and KLK7-HRP conjugate compete for the anti-KLK7 antibody binding site. Since the number of sites is limited, as more sites are occupied by KLK7 from the sample, fewer sites are left to bind KLK7-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The KLK7 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the kallikrein subfamily of serine proteases. These enzymes have diverse physiological functions and many kallikrein genes are biomarkers for cancer. The encoded protein has chymotrypsin-like activity and plays a role in the proteolysis of intercellular cohesive structures that precedes desquamation, the shedding of the outermost layer of the epidermis. The encoded protein may play a role in cancer invasion and metastasis, and increased expression of this gene is associated with unfavorable prognosis and progression of several types of cancer. Polymorphisms in this gene may play a role in the development of atopic dermatitis. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene, which is one of fifteen kallikrein subfamily members located in a gene cluster on chromosome 19. Specific for amino acid residues with aromatic side chains in the P1 position. SCCE cleaves insulin B chain at '6-Leu-|-Cys-7', '16-Tyr-|-Leu-17', '25-Phe-|-Tyr-26' and '26-Tyr-|-Thr-27'. Could play a role in the activation of precursors to inflammatory cytokines. Belongs to the peptidase S1 family. Kallikrein subfamily. 2 isoforms of the human protein are produced by alternative splicing.
