Intended Uses: This ITGalpha6 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey ITGalpha6. This ELISA kit for research use only!
Principle of the Assay: ITGalpha6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ITGalpha6 antibody and an ITGalpha6-HRP conjugate. The assay sample and buffer are incubated together with ITGalpha6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ITGalpha6 concentration since ITGalpha6 from samples and ITGalpha6-HRP conjugate compete for the anti-ITGalpha6 antibody binding site. Since the number of sites is limited, as more sites are occupied by ITGalpha6 from the sample, fewer sites are left to bind ITGalpha6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ITGalpha6 concentration in each sample is interpolated from this standard curve.

The ITGA6 protein product is the integrin alpha chain alpha 6. Integrins are integral cell-surface proteins composed of an alpha chain and a beta chain. A given chain may combine with multiple partners resulting in different integrins. For example, alpha 6 may combine with beta 4 in the integrin referred to as TSP180, or with beta 1 in the integrin VLA-6. Integrins are known to participate in cell adhesion as well as cell-surface mediated signalling. Two transcript variants encoding different isoforms have been found for this gene.
