Principle of the Assay: ITF2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ITF2 antibody and an ITF2-HRP conjugate. The assay sample and buffer are incubated together with ITF2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ITF2 concentration since ITF2 from samples and ITF2-HRP conjugate compete for the anti-ITF2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ITF2 from the sample, fewer sites are left to bind ITF2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ITF2 concentration in each sample is interpolated from this standard curve

NCBI Description

Members of the trefoil family are characterized by having at least one copy of the trefoil motif, a 40-amino acid domain that contains three conserved disulfides. They are stable secretory proteins expressed in gastrointestinal mucosa. Their functions are not defined, but they may protect the mucosa from insults, stabilize the mucus layer and affect healing of the epithelium. The encoded protein inhibits gastric acid secretion. This gene and two other related trefoil family member genes are found in a cluster on chromosome 21. [provided by RefSeq, Jul 2008]
