Bovine Insulin receptor substrate 1 ELISA Kit
Principle of the Assay: IRS1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-IRS1 antibody and an IRS1-HRP conjugate. The assay sample and buffer are incubated together with IRS1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IRS1 concentration since IRS1 from samples and IRS1-HRP conjugate compete for the anti-IRS1 antibody binding site. Since the number of sites is limited, as more sites are occupied by IRS1 from the sample, fewer sites are left to bind IRS1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IRS1 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a protein which is phosphorylated by insulin receptor tyrosine kinase. Mutations in this gene are associated with type II diabetes and susceptibility to insulin resistance. [provided by RefSeq, Nov 2009]
Uniprot Description
IRS1: an adaptor protein that is one of the major substrates of the insulin receptor kinase. Contains multiple tyrosine phosphorylation motifs that serve as docking sites for SH2-domain-containing proteins including phosphatidylinositol 3-kinase p85 subunit and GRB-2. Phosphorylated on ser/thr residues by kinases including JNK and IKK.

Product Notes
The Bovine IRS-1 irs1 (Catalog #AAA753040) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA753040 ELISA Kit recognizes Bovine IRS-1. It is sometimes possible for the material contained within the vial of "Insulin receptor substrate 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
