Intended Uses
This ELISA kit is intended for laboratory in vitro research use only, not for drug, household or other use! The Stop Solution changes the color from blue to yellow and the intensity of the color is measured at 450 nm by a spectrophotometer. In order to measure the concentration of Goat INS in the sample, this ELISA kit includes a set of calibration standards. The calibration standards are assayed at the same time as the samples and allow the operator to produce a standard curve of Optical Density versus Goat INS concentration. The concentration of Goat INS in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Introduction: ELISA is a simple and highly sensitive method of analysis that allows for simultaneous and rapid quantification of a large number of samples. The assay is based on the specific recognition of the target compound (analyte/antigen) by antibodies which bind to the compound. The antigen-antibody complex is detected and measured with the aid of an enzyme-labeled antibody or antigen. Upon addition of a non-colored reagent, the enzyme produces a color reaction where the color intensity is directly or inversely proportional to the concentration of the analyte in the sample.

NCBI Description

After removal of the precursor signal peptide, proinsulin is post-translationally cleaved into three peptides: the B chain and A chain peptides, which are covalently linked via two disulfide bonds to form insulin, and C-peptide. Binding of insulin to the insulin receptor (INSR) stimulates glucose uptake. A multitude of mutant alleles with phenotypic effects have been identified. There is a read-through gene, INS-IGF2, which overlaps with this gene at the 5' region and with the IGF2 gene at the 3' region. Alternative splicing results in multiple transcript variants.
