DL-threo-2-methylisocitrate is a substrate of isocitrate lyase 1 (ICL1). Using Michaelis-Menten nonlinear least squares fitting, kcat value was 5.24 s-1, the Km of the purified recombinant ICL1 of threonine-D(s)L(s)-isocitrate (ICA) was 188 uM. The Km of DL-threo-2-methylisocitrate (MICA) is approximately four times higher at 718 uM, and the kcat is 1.25 s-1.
