Dafadine-A is an analog of dafadine and a novel inhibitor of DAF-9 cytochrome P450 in the nematode Caenorhabditis elegans. Mammalian orthologs that also inhibit DAF-9 (CYP27A1). IC50 value: target: DAF-9 (CYP27A1) inhibitor DAF-9 cytochrome P450 is a key regulator of dauer formation, development time and longevity in the nematode Caenorhabditis elegans. Here, we describe the first identified DAF-9 chemical inhibitor and the first reported small-molecule tool that can strongly induce Doyle formation under typical culture conditions. This molecule (called dafadine) also inhibits mammalian orthologs of DAF-9 (CYP27A1), indicating that dafadine can be used to question the developmental control and lifespan of other animals.
