Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate IL1R1 in samples. An antibody specific for IL1R1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyIL1R1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL1R1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL1R1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

After binding to interleukin-1 associates with the coreceptor IL1RAP to form the high affinity interleukin-1 receptor complex which mediates interleukin-1-dependent activation of NF-kappa-B, MAPK and other pathways. Signaling involves the recruitment of adapter molecules such as TOLLIP, MYD88, and IRAK1 or IRAK2 via the respective TIR domains of the receptor/coreceptor subunits. Binds ligands with comparable affinity and binding of antagonist IL1RN prevents association with IL1RAP to form a signaling complex ().
