Background: Cell division cycle 5-like protein, also called PCDC5RP or CDC5-like, is a protein that in humans is encoded by the CDC5L gene. The CDC5L gene was mapped to chromosome 6p21 by fluorescence in situ hybridization. This DNA-binding protein involved in cell cycle control. It may act as a transcription activator. The Component of the PRP19-CDC5L complex forms an integral part of the spliceosome and is required for activating pre-mRNA splicing. The role of CDC5(Polo-like kinase) in regulating Rho1 is likely to be relevant to cytokinesis and asymmetric cell division in other organisms.

The protein encoded by this gene shares a significant similarity with Schizosaccharomyces pombe cdc5 gene product, which is a cell cycle regulator important for G2/M transition. This protein has been demonstrated to act as a positive regulator of cell cycle G2/M progression. It was also found to be an essential component of a non-snRNA spliceosome, which contains at least five additional protein factors and is required for the second catalytic step of pre-mRNA splicing. Is part of a spliceosomal 'core' complex that includes CDC5L, SF2, PRLI, SPF27, HSP7C, PRPF19/SNEV, GCN1L. Identified in the spliceosome C complex. Has been associated with aneuploidy in aging cells and in tumorigenesis. The genomic region containing the gene for CDC5L (6p21.1) is frequently amplified in osteosarcomas.
