Principle of the Assay: MBS Human IGFBP3 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IGFBP3 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human IGFBP3 coated on a 96-well plate. Standards and samples are pipetted into the wells and IGFBP3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IGFBP3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IGFBP3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm!!Background/Introduction: Insulin-like growth factor (IGF)-binding protein-3 (IGFBP-3) is a major determinant of circulating levels of the IGFs and is clinically useful for the evaluation of GH deficiency and for predicting the response to GH treatment. The circulating level of IGFBP-3 is inversely related to the risk of several common cancers, and that antiproliferative agents such as antiestrogens and retinoids act in part by up-regulating IGFBP-3 gene (IGFBP3) expression. Insulin-like growth factor-binding protein (IGFBP)-3, well characterized as the carrier of insulin-like growth factor (IGF), has been reported to have intrinsic bioactivity that is independent of IGF binding. IGFBP-3 has an IGF-independent, antiproliferative effect in undifferentiated and early differentiated but not in terminally differentiated chondrocytes. IGFBP-3 possesses both growth-inhibitory and potentiating effects on cells that are independent of IGF action and are mediated through specific. IGFBP-3 binding proteins/receptors locate at the cell membrane, cytosol, or nuclear compartments and in the extrIGFBP-3llular matrix. IGFBP3 is also located on chromosome 7.

NCBI Description

The protein encoded by this gene is a cytokine that regulates T and natural killer cell activation and proliferation. This cytokine and interleukine 2 share many biological activities. They are found to bind common hematopoietin receptor subunits, and may compete for the same receptor, and thus negatively regulate each other's activity. The number of CD8+ memory cells is shown to be controlled by a balance between this cytokine and IL2. This cytokine induces the activation of JAK kinases, as well as the phosphorylation and activation of transcription activators STAT3, STAT5, and STAT6. Studies of the mouse counterpart suggested that this cytokine may increase the expression of apoptosis inhibitor BCL2L1/BCL-x(L), possibly through the transcription activation activity of STAT6, and thus prevent apoptosis. Alternatively spliced transcript variants of this gene have been reported. [provided by RefSeq, Feb 2011]

Uniprot Description

IL15: Cytokine that stimulates the proliferation of T- lymphocytes. Stimulation by IL-15 requires interaction of IL-15 with components of IL-2R, including IL-2R beta and probably IL-2R gamma but not IL-2R alpha. Belongs to the IL-15/IL-21 family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human IL15 il15 (Catalog #AAA824843) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA824843 ELISA Kit recognizes Human IL15. AAA Biotech's IL-15 can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (SE). Researchers should empirically determine the suitability of the IL15 il15 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IL-15, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
