Principle of the Assay: IL-16 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-IL-16 antibody and an IL-16-HRP conjugate. The assay sample and buffer are incubated together with IL-16-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-16 concentration since IL-16 from samples and IL-16-HRP conjugate compete for the anti-IL-16 antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-16 from the sample, fewer sites are left to bind IL-16-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-16 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a pleiotropic cytokine that functions as a chemoattractant, a modulator of T cell activation, and an inhibitor of HIV replication. The signaling process of this cytokine is mediated by CD4. The product of this gene undergoes proteolytic processing, which is found to yield two functional proteins. The cytokine function is exclusively attributed to the secreted C-terminal peptide, while the N-terminal product may play a role in cell cycle control. Caspase 3 is reported to be involved in the proteolytic processing of this protein. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Feb 2010]
