Porcine Interferon lambda-3 ELISA Kit

Principle of the Assay: IFNlambda3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IFNlambda3 antibody and an IFNlambda3-HRP conjugate. The assay sample and buffer are incubated together with IFNlambda3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IFNlambda3 concentration since IFNlambda3 from samples and IFNlambda3-HRP conjugate compete for the anti-IFNlambda3 antibody binding site. Since the number of sites is limited, as more sites are occupied by IFNlambda3 from the sample, fewer sites are left to bind IFNlambda3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IFNlambda3 concentration in each sample is interpolated from this standard curve.

This gene encodes a cytokine distantly related to type I interferons and the IL-10 family. This gene, interleukin 28A (IL28A), and interleukin 29 (IL29) are three closely related cytokine genes that form a cytokine gene cluster on a chromosomal region mapped to 19q13. Expression of the cytokines encoded by the three genes can be induced by viral infection. All three cytokines have been shown to interact with a heterodimeric class II cytokine receptor that consists of interleukin 10 receptor, beta (IL10RB) and interleukin 28 receptor, alpha (IL28RA). Seems not to be essential for early virus-activated host defense in vaginal infection, but plays an important role in Toll-like receptor (TLR)-induced antiviral defense. Plays a significant role in the antiviral immune defense in the intestinal epithelium. Exerts an immunomodulatory effect by up-regulating MHC class I antigen expression.
