Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate IFNAR1 in samples. An antibody specific for IFNAR1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyIFNAR1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IFNAR1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IFNAR1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

Component of the receptor for type I interferons, including interferons alpha, IFNB1 and IFNW1. Functions in general as heterodimer with IFNAR2. Type I interferon binding activates the JAK-STAT signaling cascade, and triggers tyrosine phosphorylation of a number of proteins including JAKs, TYK2, STAT proteins and the IFNR alpha- and beta-subunits themselves (). Can form an active IFNB1 receptor by itself and activate a signaling cascade that does not involve activation of the JAK-STAT pathway ().
