INTENDED USE This ICAM-3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Goat ICAM-3. This ELISA kit for research use only!

PRINCIPLE OF THE ASSAY ICAM-3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ICAM-3 antibody and an ICAM-3-HRP conjugate. The assay sample and buffer are incubated together with ICAM-3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ICAM-3 concentration since ICAM-3 from samples and ICAM-3-HRP conjugate compete for the anti-ICAM-3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ICAM-3 from the sample, fewer sites are left to bind ICAM-3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ICAM-3 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the intercellular adhesion molecule (ICAM) family. All ICAM proteins are type I transmembrane glycoproteins, contain 2-9 immunoglobulin-like C2-type domains, and bind to the leukocyte adhesion LFA-1 protein. This protein is constitutively and abundantly expressed by all leucocytes and may be the most important ligand for LFA-1 in the initiation of the immune response.
