Principle of the assay: HYOU1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HYOU1 antibody and an HYOU1-HRP conjugate. The assay sample and buffer are incubated together with HYOU1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HYOU1 concentration since HYOU1 from samples and HYOU1-HRP conjugate compete for the anti-HYOU1 antibody binding site. Since the number of sites is limited, as more sites are occupied by HYOU1 from the sample, fewer sites are left to bind HYOU1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HYOU1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the heat shock protein 70 family. This gene uses alternative transcription start sites. A cis-acting segment found in the 5' UTR is involved in stress-dependent induction, resulting in the accumulation of this protein in the endoplasmic reticulum (ER) under hypoxic conditions. The protein encoded by this gene is thought to play an important role in protein folding and secretion in the ER. Since suppression of the protein is associated with accelerated apoptosis, it is also suggested to have an important cytoprotective role in hypoxia-induced cellular perturbation. This protein has been shown to be up-regulated in tumors, especially in breast tumors, and thus it is associated with tumor invasiveness. This gene also has an alternative translation initiation site, resulting in a protein that lacks the N-terminal signal peptide. This signal peptide-lacking protein, which is only 3 amino acids shorter than the mature protein in the ER, is thought to have a housekeeping function in the cytosol. In rat, this protein localizes to both the ER by a carboxy-terminal peptide sequence and to mitochondria by an amino-terminal targeting signal.
