Principle of the Assay: The PKC theta (Phospho-Thr538) Cell-Based ELISA Kit is a convenient, lysate- free, high throughput and sensitive assay kit that can monitor PKC theta protein phosphorylation and expression profile in cells. The kit can be used for measuring the relative amounts of phosphorylated PKC theta in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on PKC theta phosphorylation.

Background/Introduction: The Colorimetric Cell-Based ELISA Kit allows for the detection of various target proteins and the effects that certain stimulation conditions have on target protein expression in different cell lines. Qualitative determination of target protein concentration is achieved by an indirect ELISA format. In essence, the target protein is captured by target-specific primary (1 degree) antibodies while the HRP-conjugated secondary (2 degree) antibodies bind the Fc region of the 1 degree antibody. Through this binding, the HRP enzyme conjugated to the 2 degree antibody can catalyze a colorimetric reaction upon substrate addition. Due to the qualitative nature of the Cell-Based ELISA, multiple normalization methods are described: 1) a monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values. 2) Following the colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method is used to determine cell density. After staining, the results can be analyzed by normalizing the absorbance values to cell amounts, by which the plating difference can be adjusted. 3) If a phosphorylated target is being detected, an antibody against the non- phosphorylated counterpart will be provided for normalization purposes. The absorbance values obtained for the non-phosphorylated target can be used to normalize the absorbance values for the phosphorylated target.

NCBI Description

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role. The protein encoded by this gene is one of the PKC family members. It is a calcium-independent and phospholipid-dependent protein kinase. This kinase is important for T-cell activation. It is required for the activation of the transcription factors NF-kappaB and AP-1, and may link the T cell receptor (TCR) signaling complex to the activation of the transcription factors. Uniprot Description

PKCT: an AGC kinase of the PKC family. A novel PKC: Ca2+ independent but still regulated by PS, DAG and phorbol esters. Contains 2 zinc-dependent phorbol-ester and DAG binding domains. Expressed in skeletal muscle and hematopoietic cells. May play a role in T cell receptor signaling and insulin resistance. Required for TCR-induced NF-kappaB activation in mature T lymphocytes.

Product Notes

The Human, Mouse, Rat PKC theta prkcq (Catalog #AAA9501215) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9501215 ELISA Kit recognizes Human, Mouse, Rat PKC theta. It is sometimes possible for the material contained within the vial of "PKC theta, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
