Specificity
The protein detected by this kit corresponds to Swiss-Prot Acc. Q13541 and Entrez-Gene Id 1978. This assay kit contains 2 highly specific and selective antibodies that recognize the phospho-Thr37/Thr46 epitope and a distinct epitope on 4EBP1. These antibodies recognize 4EBP1 (T37/T46) of human, mouse, rat and monkey origin. Other species should be tested on a case-by-case basis.

Testing Data

(A431 cells were seeded at 25,000 cells/well and cultured overnight at 37 degree C, 5% CO2, in DMEM with 10% FBS. Cells were then treated with PP242 for 3 h at 37 degree C. The media was removed from the wells, and cells were lysed with 50 uL/well of 1X Lysis Buffer #2 containing 1 mM NaF and 2 mM Na3VO4, on an orbital shaker (400 rpm) for 30 min at room temperature. The lysates were then analyzed for phospho-4EBP1 (T37/T46) using the two-plate assay protocol. The TR-FRET signal was recorded at 665 and 615 nm (EnVision lamp excitation) after a 4 h incubation period at room temperature.)

NCBI Description

This gene encodes one member of a family of translation repressor proteins. The protein directly interacts with eukaryotic translation initiation factor 4E (eIF4E), which is a limiting component of the multisubunit complex that recruits 40S ribosomal subunits to the 5' end of mRNAs. Interaction of this protein with eIF4E inhibits complex assembly and represses translation. This protein is phosphorylated in response to various signals including UV irradiation and insulin signaling, resulting in its dissociation from eIF4E and activation of mRNA translation. 4E-BP1: binds to eIF4E, preventing its assembly into the EIF4F complex and inhibiting cap-dependent translation. Phosphorylation of 4E-BP1 disrupts this binding, activating cap-dependent translation. Mediates the regulation of protein translation by hormones, growth factors and other stimuli that signal through the PI3 kinase pathway.
