Principle of the Assay: This kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. The purified anti-Vimentin antibody was pre-coated onto 96-well plates. And the HRP conjugated anti-Vimentin antibody was used as detection antibodies. The standards, test samples and HRP conjugated detection antibody were added to the wells subsequently, mixed and incubated, then, unbound conjugates were washed away with wash buffer. TMB substrates (A & B) were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Vimentin amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Vimentin can be calculated.

Background/Introduction: Vimentin is a type III intermediate filament (IF) protein that is expressed in mesenchymal cells. IF proteins are found in all metazoan cells as well as bacteria. All IF proteins are expressed in a highly developmentally-regulated fashion; vimentin is the major cytoskeletal component of mesenchymal cells. Because of this, vimentin is often used as a marker of mesenchymally-derived cells or cells undergoing an epithelial-to-mesenchymal transition (EMT) during both normal development and metastatic progression. It plays a significant role in supporting and anchoring the position of the organelles in the cytosol, and it has been used as a sarcoma tumor marker to identify mesenchyme.

NCBI Description

This gene encodes a member of the intermediate filament family. Intermediate filamentents, along with microtubules and actin microfilaments, make up the cytoskeleton. The protein encoded by this gene is responsible for maintaining cell shape, integrity of the cytoplasm, and stabilizing cytoskeletal interactions. It is also involved in the immune response, and controls the transport of low-density lipoprotein (LDL)-derived cholesterol from a lysosome to the site of esterification. It functions as an organizer of a number of critical proteins involved in attachment, migration, and cell signaling. Mutations in this gene causes a dominant, pulverulent cataract.[provided by RefSeq, Jun 2009]

Uniprot Description

Function: Vimentins are class-III intermediate filaments found in various non-epithelial cells, especially mesenchymal cells. Vimentin is attached to the nucleus, endoplasmic reticulum, and mitochondria, either laterally or terminally. Ref.37Involved with LARP6 in the stabilization of type I collagen mRNAs for CO1A1 and CO1A2. Ref.37

Subunit structure: Homopolymer assembled from elementary dimers. Interacts with HCV core protein. Interacts with LGSN and SYNM. Interacts (via rod region) with PLEC (via CH 1 domain)

By similarity. Interacts with SLC6A4. Interacts with STK33. Interacts with LARP6. Interacts with RAB8B

By similarity. Ref.23 Ref.27 Ref.32 Ref.37 Ref.42

Subcellular location: Cytoplasm Ref.39.

Tissue specificity: Highly expressed in fibroblasts, some expression in T- and B-lymphocytes, and little or no expression in Burkitt's lymphoma cell lines. Expressed in many hormone-independent mammary carcinoma cell lines. Ref.12 Ref.17

Domain: The central alpha-helical coiled-coil rod region mediates elementary homodimerization. Ref.42

Post-translational modification: Filament disassembly during mitosis is promoted by phosphorylation at Ser-55 as well as by nestin

By similarity. One of the most prominent phosphoproteins in various cells of mesenchymal origin. Phosphorylation is enhanced during cell division, at which time vimentin filaments are significantly reorganized. Phosphorylation by PKN1 inhibits the formation of filaments. Phosphorylated at Ser-56 by CDK5 during neutrophil secretion in the cytoplasm. Phosphorylated by STK33. Ref.14 Ref.19 Ref.20 Ref.21 Ref.27 Ref.39

Sequence similarities: Belongs to the intermediate filament family.

Sequence caution: The sequence BAB71275.1 differs from that shown. Reason: Intron retention.

Product Notes

The Human VIM vim (Catalog #AAA355298) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355298 ELISA Kit recognizes Human VIM. AAA Biotech's Vimentin can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of Vimentin in Human serum, plasma, urine, cell culture supernatant or tissue samples. Researchers should empirically determine the suitability of the VIM vim for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Vimentin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
