Intended Uses: This YARS ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human YARS. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: YARS ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-YARS antibody and an YARS-HRP conjugate. The assay sample and buffer are incubated together with YARS-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the YARS concentration since YARS from samples and YARS-HRP conjugate compete for the anti-YARS antibody binding site. Since the number of sites is limited, as more sites are occupied by YARS from the sample, fewer sites are left to bind YARS-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The YARS concentration in each sample is interpolated from this standard curve.

NCBI Description

Aminoacyl-tRNA synthetases catalyze the aminoacylation of tRNA by their cognate amino acid. Because of their central role in linking amino acids with nucleotide triplets contained in tRNAs, aminoacyl-tRNA synthetases are thought to be among the first proteins that appeared in evolution. Tyrosyl-tRNA synthetase belongs to the class I tRNA synthetase family. Cytokine activities have also been observed for the human tyrosyl-tRNA synthetase, after it is split into two parts, an N-terminal fragment that harbors the catalytic site and a C-terminal fragment found only in the mammalian enzyme. The N-terminal fragment is an interleukin-8-like cytokine, whereas the released C-terminal fragment is an EMAP II-like cytokine. [provided by RefSeq, Jul 2008]

YARS: Catalyzes the attachment of tyrosine to tRNA(Tyr) in a two-step reaction: tyrosine is first activated by ATP to form Tyr- AMP and then transferred to the acceptor end of tRNA(Tyr). Defects in YARS are the cause of Charcot-Marie-Tooth disease dominant intermediate type C (CMTDIC). CMTDIC is a form of Charcot-Marie-Tooth disease characterized by clinical and pathologic features intermediate between demyelinating and axonal peripheral neuropathies, and motor median nerve conduction velocities ranging from 25 to 45 m/sec. Belongs to the class-I aminoacyl-tRNA synthetase family.
