Assay Procedure Summary
1. Prepare all reagents, samples and standards
2. Add 25ul standard or sample to each well. Incubate 1 hour at 37 degree C
3. Aspirate and add 25ul prepared Detection Reagent A. Incubate 1 hour at 37 degree C
4. Aspirate and wash 3 times
5. Add 25ul prepared Detection Reagent B. Incubate 30 minutes at 37 degree C
6. Aspirate and wash 5 times
7. Add 25ul Substrate Solution. Incubate 10-20 minutes at 37 degree C
8. Add 20ul Stop Solution. Read at 450nm immediately.
Test Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Lactoperoxidase (LPO). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Lactoperoxidase (LPO). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Lactoperoxidase (LPO), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Lactoperoxidase (LPO) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Preparation and Storage
Store at 4 degree C for 1 month/ -20 degree C for long-term (within shelf life).
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

NCBI Description

The protein encoded by this gene is the tropomyosin-binding subunit of the troponin complex, which is located on the thin filament of striated muscles and regulates muscle contraction in response to alterations in intracellular calcium ion concentration. Mutations in this gene have been associated with familial hypertrophic cardiomyopathy as well as with dilated cardiomyopathy. Transcripts for this gene undergo alternative splicing that results in many tissue-specific isoforms, however, the full-length nature of some of these variants has not yet been determined. TNNT2: Troponin T is the tropomyosin-binding subunit of troponin, the thin filament regulatory complex which confers calcium-sensitivity to striated muscle actomyosin ATPase activity. Heart. The fetal heart shows a greater expression in the atrium than in the ventricle, while the adult heart shows a greater expression in the ventricle than in the atrium. Isoform 6 predominates in normal adult heart. Isoforms 1, 7 and 8 are expressed in fetal heart. Isoform 7 is also expressed in failing adult heart. Belongs to the troponin T family. 11 isoforms of the human protein are produced by alternative splicing.
