Intended Uses: This TPM1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human TPM1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: TPM1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-TPM1 antibody and an TPM1-HRP conjugate. The assay sample and buffer are incubated together with TPM1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TPM1 concentration since TPM1 from samples and TPM1-HRP conjugate compete for the anti-TPM1 antibody binding site. Since the number of sites is limited, as more sites are occupied by TPM1 from the sample, fewer sites are left to bind TPM1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TPM1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is a member of the tropomyosin family of highly conserved, widely distributed actin-binding proteins involved in the contractile system of striated and smooth muscles and the cytoskeleton of non-muscle cells. Tropomyosin is composed of two alpha-helical chains arranged as a coiled-coil. It is polymerized end to end along the two grooves of actin filaments and provides stability to the filaments. The encoded protein is one type of alpha helical chain that forms the predominant tropomyosin of striated muscle, where it also functions in association with the troponin complex to regulate the calcium-dependent interaction of actin and myosin during muscle contraction. In smooth muscle and non-muscle cells, alternatively spliced transcript variants encoding a range of isoforms have been described. Mutations in this gene are associated with type 3 familial hypertrophic cardiomyopathy. [provided by RefSeq, Jul 2008]

Plays a central role, in association with the troponin complex, in the calcium dependent regulation of vertebrate striated muscle contraction. Smooth muscle contraction is regulated by interaction with caldesmon. In non-muscle cells is implicated in stabilizing cytoskeleton actin filaments. Defects in TPM3 are a cause of nemaline myopathy type 1 (NEM1). Three alternatively spliced isoforms have been described.
