* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: TNIP2 binds to the C-terminal zinc finger domain of A20 (TNFAIP3) and is involved in activation of the ERK MAP kinase pathway in various cell types. By yeast 2-hybrid screening of a mouse fibrosarcoma cDNA library using A20 as bait, followed by database analysis, Van Huffel et al. (2001) identified human TNIP2, which they called ABIN2. The mouse and human proteins share 76% amino acid identity. Northern blot analysis of mouse tissues revealed ubiquitous expression, with highest expression in kidney and weakest expression in skeletal muscle. Western blot analysis revealed a 50-kD endogenous Abin2 protein in mouse macrophages and fibrosarcoma cells.

Forms a ternary complex with NFKB1 and MAP3K8 but appears to function upstream of MAP3K8 in the TLR4 signaling pathway that regulates MAP3K8 activation. Involved in activation of the MEK/ERK signaling pathway during innate immune response; this function seems to be stimulus- and cell type specific. Required for stability of MAP3K8. Involved in regulation of apoptosis in endothelial cells; promotes TEK agonist-stimulated endothelial survival. May act as transcriptional coactivator when translocated to the nucleus. Enhances CHUK-mediated NF-kappa-B activation involving NF-kappa-B p50-p65 and p50-c-Rel complexes. 2 isoforms of the human protein are produced by alternative splicing.
