Principle of the Assay: The Human TGFBR3 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human TGFBR3 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human TGFBR3 coated on a 96-well plate. Standards and samples are pipetted into the wells and TGFBR3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human TGFBR3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TGFBR3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Betaglycan also known as Transforming growth factor beta receptor III (TGFBR3), is a cell- surfacechondroitin sulfate / heparan sulfate proteoglycan >300 kDa in molecular weight. Betaglycan binds to various members of the TGF-beta superfamily of ligands via its core protein, and bFGF via its heparan sulfate chains. It is not involved directly in TGF-beta signal transduction but by binding to various member of the TGF-beta superfamily at the cell surface it acts as a reservoir of ligand for TGF-beta receptors. By cDNA array and immunohistochemistry analyses, it is found that TGFBR3 expression is lost in most breast cancers examined in association with loss of heterozygosity of the TGFBR3 locus. TGFBR3 expression decreased during breast cancer progression, and low levels predicted decreased recurrence-free survival in patients. Loss of TGFBR3 through allelic imbalance is a frequent genetic event during breast cancer development that increases metastatic potential. Also, TGFBR3 can localize transformation in the heart and play an essential, nonredundant role in TGF-beta signaling.

NCBI Description

This locus encodes the transforming growth factor (TGF)-beta type III receptor. The encoded receptor is a membrane proteoglycan that often functions as a co-receptor with other TGF-beta receptor superfamily members. Ectodomain shedding produces soluble TGFBR3, which may inhibit TGFB signaling. Decreased expression of this receptor has been observed in various cancers. Alternatively spliced transcript variants encoding different isoforms have been identified for this gene.[provided by RefSeq, Sep 2010]

Uniprot Description

TGFBR3: Binds to TGF-beta. Could be involved in capturing and retaining TGF-beta for presentation to the signaling receptors. Interacts with TCTEX1D4. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human TGFBR3 tgfbr3 (Catalog #AAA8248407) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8248407 ELISA Kit recognizes Human TGFBR3. It is sometimes possible for the material contained within the vial of "TGFBR3, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
