Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for SPON2 elisa kit
Description: This Human Spondin-2 (SPON2) ELISA Kit employs a two-site sandwich ELISA to quantitate SPON2 in samples. An antibody specific for SPON2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPON2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPON2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPON2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: The full-length SPON2 cDNA encodes a 331-amino acid protein with a domain organization similar to those of zebrafish mindin-1/mindin-2 and F-spondin (SPON1): a hydrophobic signal sequence in the N terminus, an FS1 domain, an FS2 domain, and a thrombospondin type I repeat. The mouse mindin protein, which is 85% identical to the human protein, is a conserved member of a family of secreted extracellular matrix proteins. RNA blot analysis detected expression in a variety of tissues, with abundant expression in lung and lymphoid tissues. Immunoblot blot analysis showed expression of a 42-kD protein under reducing conditions and expression of dimers and oligomers in a concentration-dependent manner under nonreducing conditions.

Uniprot Description

SPON2: Cell adhesion protein that promotes adhesion and outgrowth of hippocampal embryonic neurons. Binds directly to bacteria and their components and functions as an opsonin for macrophage phagocytosis of bacteria. Essential in the initiation of the innate immune response and represents a unique pattern- recognition molecule in the ECM for microbial pathogens. Binds bacterial lipopolysaccharide (LPS).

Protein type: Secreted, signal peptide; Secreted

Chromosomal Location of Human Ortholog: 4p16.3

Cellular Component: extracellular space; proteinaceous extracellular matrix

Molecular Function: lipopolysaccharide binding; metal ion binding; antigen binding

Biological Process: induction of bacterial agglutination; mast cell mediated immunity; axon guidance; innate immune response; positive regulation of interleukin-6 production; opsonization; cell adhesion; positive regulation of tumor necrosis factor production; defense response to fungus; defense response to virus

Product Notes

The Human SPON2 spon2 (Catalog #AAA9714005) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9714005 ELISA Kit recognizes Human SPON2. It is sometimes possible for the material contained within the vial of "Spondin-2 (SPON2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
