Intended Uses: This SERPINB10 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human SERPINB10. This ELISA kit for research use only!

Principle of the Assay: SERPINB10 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-SERPINB10 antibody and an SERPINB10-HRP conjugate. The assay sample and buffer are incubated together with SERPINB10-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SERPINB10 concentration since SERPINB10 from samples and SERPINB10-HRP conjugate compete for the anti-SERPINB10 antibody binding site. Since the number of sites is limited, as more sites are occupied by SERPINB10 from the sample, fewer sites are left to bind SERPINB10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SERPINB10 concentration in each sample is interpolated from this standard curve.

NCBI Description

The superfamily of high molecular weight serine proteinase inhibitors (serpins) regulate a diverse set of intracellular and extracellular processes such as complement activation, fibrinolysis, coagulation, cellular differentiation, tumor suppression, apoptosis, and cell migration. Serpins are characterized by a well-conserved tertiary structure that consists of 3 beta sheets and 8 or 9 alpha helices (Huber and Carrell, 1989 [PubMed 2690952]). A critical portion of the molecule, the reactive center loop connects beta sheets A and C. Protease inhibitor-10 (PI10; SERPINB10) is a member of the ov-serpin subfamily, which, relative to the archetypal serpin PI1 (MIM 107400), is characterized by a high degree of homology to chicken ovalbumin, lack of N- and C-terminal extensions, absence of a signal peptide, and a serine rather than an asparagine residue at the penultimate position (summary by Bartuski et al., 1997 [PubMed 9268635]).[supplied by OMIM, Jan 2010]

Uniprot Description

Protein type: Inhibitor

Chromosomal Location of Human Ortholog: 18q21.3

Product Notes

The Human SERPINB10 serpinb10 (Catalog #AAA7237593) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7237593 ELISA Kit recognizes Human SERPINB10. It is sometimes possible for the material contained within the vial of "Serpin B10 (SERPINB10), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
