Human S phase kinase associated protein 1 (SKP1) ELISA Kit
Principle of the Assay: SKP1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-SKP1 antibody and an SKP1-HRP conjugate. The assay sample and buffer are incubated together with SKP1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SKP1 concentration since SKP1 from samples and SKP1-HRP conjugate compete for the anti-SKP1 antibody binding site. Since the number of sites is limited, as more sites are occupied by SKP1 from the sample, fewer sites are left to bind SKP1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SKP1 concentration in each sample is interpolated from this standard curve.

This gene encodes a component of SCF complexes, which are composed of this protein, cullin 1, a ring-box protein, and one member of the F-box family of proteins. This protein binds directly to the F-box motif found in F-box proteins. SCF complexes are involved in the regulated ubiquitination of specific protein substrates, which targets them for degradation by the proteosome. Specific F-box proteins recognize different target protein(s), and many specific SCF substrates have been identified including regulators of cell cycle progression and development. Studies have also characterized the protein as an RNA polymerase II elongation factor. Alternative splicing of this gene results in two transcript variants.
