Principle of the Assay: Human Runt-related Transcription Factor 3 (RUNX3) ELISA Kit employs a two-site sandwich ELISA to quantitate RUNX3 in samples. An antibody specific for RUNX3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any RUNX3 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human RUNX3 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of RUNX3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background/Introduction: RUNX3 is a member of the runt domain-containing family of transcription factors. A heterodimer of this protein and a beta subunit forms a complex that binds to the core DNA sequence 5'-PYGPYGGT-3' found in a number of enhancers and promoters, and can either activate or suppress transcription. It also interacts with other transcriptionally silenced in cancer. Multiple transcript variants encoding different isoforms have been found for this gene. Runx3 null mouse gastric mucosa exhibits hyperplasia due to stimulated proliferation and suppressed apoptosis in epithelial cells, and the cells are resistant to TGF-beta stimulation.

This gene encodes a member of the runt domain-containing family of transcription factors. A heterodimer of this protein and a beta subunit forms a complex that binds to the core DNA sequence 5'-PYGPYGGT-3' found in a number of enhancers and promoters, and can either activate or suppress transcription. It also interacts with other transcription factors. It functions as a tumor suppressor, and the gene is frequently deleted or transcriptionally silenced in cancer. AML2: CBF binds to the core site, 5'-PYGPYGGT-3', of a number of enhancers and promoters, including murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, lck, IL-3 and GM-CSF promoters. Heterodimer of an alpha and a beta subunit. The alpha subunit binds DNA as a monomer and through the Runt domain. DNA- binding is increased by heterodimerization. Interacts with TLE1 and SUV39H1. The tyrosine phosphorylated form (via runt domain) interacts with SRC (via protein kinase domain). Interacts with FYN and LCK. 2 isoforms of the human protein are produced by alternative splicing.
