Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Relaxin (RLN) ELISA Kit employs a two-site sandwich ELISA to quantitate RLN1 in samples. An antibody specific for RLN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any RLN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RLN1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RLN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Relaxin is a peptide hormone produced by the corpora lutea of ovaries during pregnancy in many mammalian species, including man.The secretion of the hormone into the blood stream just before parturition results in a marked softening and lengthening of the pubic symphysis and a softening of the cervix, which facilitates the birth process. By inhibiting uterine contractions, relaxin may influence the timing of parturition. Like insulin, relaxin consists of two peptide chains, A and B, covalently linked by disulfide bonds. By further analogy to insulin, the two peptides are synthesized as a single-chain precursor polypeptide with the B chain at the NH2-terminus.

NCBI Description

Relaxins are known endocrine and autocrine/paracrine hormones, belonging to the insulin gene superfamily. In humans there are three non-allelic relaxin genes, RLN1, RLN2 and RLN3, where RLN1 and RLN2 share high sequence homology. The protein encoded by this gene is synthesized as a single-chain polypeptide but the active form consists of an A chain and a B chain linked by disulfide bonds. Relaxin is produced by the ovary, and targets the mammalian reproductive system to ripen the cervix, elongate the pubic symphysis and inhibit uterine contraction. It may have additional roles in enhancing sperm motility, regulating blood pressure, controlling heart rate and releasing oxytocin and vasopressin. [provided by RefSeq, Jan 2013]
