Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PIF/DCD elisa kit
Description: This Human Proteolysis Inducing Factor/Dermcidin (PIF/DCD) ELISA Kit employs a two-site sandwich ELISA to quantitate DCD in samples. An antibody specific for DCD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any DCD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DCD is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DCD bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Dermcidin is a secreted protein that is subsequently processed into mature peptides of distinct biological activities. The C-terminal peptide is constitutively expressed in sweat and has antibacterial and antifungal activities. The N-terminal peptide, also known as diffusible survival evasion peptide, promotes neural cell survival under conditions of severe oxidative stress. A glycosylated form of the N-terminal peptide may be associated with cachexia (muscle wasting) in cancer patients.High performance liquid chromatography, mass spectrometry, and microsequence analysis of sweat showed a processed 47-amino acid DCD peptide with a calculated mass of 4.7 kD, compared with the expected 9.3 kD of full-length DCD without the signal peptide.DCD expression in skin, melanocytic nevus, and cutaneous melanoma tissue, but not in any other organ systems tested.

NCBI Description

This antimicrobial gene encodes a secreted protein that is subsequently processed into mature peptides of distinct biological activities. The C-terminal peptide is constitutively expressed in sweat and has antibacterial and antifungal activities. The N-terminal peptide, also known as diffusible survival evasion peptide, promotes neural cell survival under conditions of severe oxidative stress. A glycosylated form of the N-terminal peptide may be associated with cachexia (muscle wasting) in cancer patients. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Oct 2014]

Uniprot Description

DCD: DCD-1 displays antimicrobial activity thereby limiting skin infection by potential pathogens in the first few hours after bacterial colonization. Highly effective against E.coli, E.faecalis, S.aureus and C.albicans. Optimal pH and salt concentration resemble the conditions in sweat. Also exhibits proteolytic activity. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Secreted; Secreted, signal peptide; EC 3.4.-.-

Chromosomal Location of Human Ortholog: 12q13.1

Cellular Component: extracellular region

Molecular Function: peptidase activity

Biological Process: killing of cells of another organism; defense response to bacterium; proteolysis; defense response to fungus

Product Notes

The Human PIF/DCD dcd (Catalog #AAA9716072) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9716072 ELISA Kit recognizes Human PIF/DCD. It is sometimes possible for the material contained within the vial of "Proteolysis Inducing Factor/Dermcidin (PIF/DCD), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
