Introduction: Human pregnancy-specific beta-1 glycoproteins (SP1) are a group of closely related placental glycoproteins and members of the immunoglobulin superfamily. These proteins can be produced in human intestinal tissue, T lymphocytes and male reproductive tract. However, they are mainly synthesized in the syncytiotrophoblast of placenta and secreted into the maternal circulation in increasing amounts as gestation progresses. In males and non-pregnant females, circulating levels of the protein are less than 3 ug/ml. SP1 levels have been suggested to correlate with certain obstetrical complications.

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific for SP1 antibody has been pre-coated onto a microplate. A competitive inhibition reaction is launched between HRP labeled SP1 and unlabeled SP1 (Standards or samples) with the antibody specific for SP1. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of SP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The human placenta is a multihormonal endocrine organ that produces hormones, enzymes, and other molecules that support fetal survival and development. Pregnancy-specific beta-1-glycoprotein (PSBG, PSG) is a major product of the syncytiotrophoblast, reaching concentrations of 100 to 290 mg/l at term in the serum of pregnant women (Horne et al., 1976 [PubMed 971765]). PSG is a member of the immunoglobulin (Ig) superfamily (Watanabe and Chou, 1988 [PubMed 3257488]; Streydio et al., 1988 [PubMed 3260773]).[supplied by OMIM, Oct 2009]
