Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PTN elisa kit
Description: This Human Pleiotrophin (PTN) ELISA Kit employs a two-site sandwich ELISA to quantitate PTN in samples. An antibody specific for PTN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PTN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PTN is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTN bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Pleiotrophin was initially recognized as a neurite outgrowth-promoting factor present in rat brain around birth and as a mitogen toward fibroblasts isolated from bovine uterus tissue. Together with midkine these growth-factors constitute a family of secreted heparin-binding proteins now known as the Neurite Growth-promoting Factor (NEGF) family. During embryonic and early postnatal development, Pleiotrophin is expressed in the central and peripheral nervous system and also in several non-neural tissues, notably lung, kidney, gut and bone. Pleiotrophin is also expressed by several tumor cells and is thought to be involved in tumor angiogenesis. In the adult central nervous system. Pleiotrophin is expressed in an activity-dependent manner in the hippocampus, where it can suppress long term potentiation induction.

NCBI Description

The protein encoded by this gene is a secreted heparin-binding growth factor. The protein has significant roles in cell growth and survival, cell migration, angiogenesis and tumorigenesis. Alternative splicing and the use of alternative promoters results in multiple transcript variants. [provided by RefSeq, Oct 2016]

Uniprot Description

PTN: Secreted growth factor that induces neurite outgrowth and which is mitogenic for fibroblasts, epithelial, and endothelial cells. Binds anaplastic lymphoma kinase (ALK) which induces MAPK pathway activation, an important step in the anti- apoptotic signaling of PTN and regulation of cell proliferation. Belongs to the pleiotrophin family.

Protein type: Secreted, signal peptide; Secreted

Chromosomal Location of Human Ortholog: 7q33

Cellular Component: extracellular space; endoplasmic reticulum

Molecular Function: heparin binding; growth factor activity; protein phosphatase inhibitor activity

Biological Process: nervous system development; positive regulation of cell division; positive regulation of cell proliferation; negative regulation of catalytic activity; learning; transmembrane receptor protein tyrosine phosphatase signaling pathway; bone mineralization

Product Notes

The Human PTN ptn (Catalog #AAA9716252) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9716252 ELISA Kit recognizes Human PTN. It is sometimes possible for the material contained within the vial of "Pleiotrophin (PTN), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
