Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Related Product Information for MAdCAM1 elisa kit
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Mucosal Addressin Cell Adhesion Molecule 1 (MAdCAM1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Mucosal Addressin Cell Adhesion Molecule 1 (MAdCAM1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Mucosal Addressin Cell Adhesion Molecule 1 (MAdCAM1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Mucosal Addressin Cell Adhesion Molecule 1 (MAdCAM1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

The protein encoded by this gene is an endothelial cell adhesion molecule that interacts preferentially with the leukocyte beta7 integrin LPAM-1 (alpha4beta7), L-selectin, and VLA-4 (alpha4beta1) on myeloid cells to direct leukocytes into mucosal and inflamed tissues. It is a member of the immunoglobulin family and is similar to ICAM1 and VCAM1. At least seven alternatively spliced transcripts encoding different protein isoforms have been found for this gene, but the full-length nature of some variants has not been determined. [provided by RefSeq, Jul 2008]

Uniprot Description

MADCAM1: Cell adhesion leukocyte receptor expressed by mucosal venules, helps to direct lymphocyte traffic into mucosal tissues including the Peyer patches and the intestinal lamina propria. It can bind both integrin alpha-4/beta-7 and L-selectin, regulating both the passage and retention of leukocytes. Isoform 2, lacking the mucin-like domain, may be specialized in supporting integrin alpha-4/beta-7-dependent adhesion strengthening, independent of L- selectin binding. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Motility/polarity/chemotaxis; Membrane protein, integral

Chromosomal Location of Human Ortholog: 19p13.3

Cellular Component: membrane; plasma membrane; integral to membrane

Biological Process: integrin-mediated signaling pathway; regulation of immune response; extracellular matrix organization and biogenesis; positive regulation of leukocyte migration; cell-matrix adhesion; signal transduction; receptor clustering; keratinocyte differentiation; heterotypic cell-cell adhesion; immune response; cell adhesion; leukocyte tethering or rolling; aging

Product Notes

The Human MAdCAM1 madcam1 (Catalog #AAA2021711) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2021711 ELISA Kit recognizes Human MAdCAM1. It is sometimes possible for the material contained within the vial of "Mucosal Addressin Cell Adhesion Molecule 1 (MAdCAM1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
