Principle of the Assay: Human Nuclear Receptor Corepressor 2 (NCOR2) ELISA Kit employs a two-site sandwich ELISA to quantitate NCOR2 in samples. An antibody specific for NCOR2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NCOR2 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human NCOR2 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of NCOR2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

In addition, NCOR2 appears to recruit histone deacetylases to DNA promoter regions. Hence NCOR2 assists nuclear receptors in the down regulation of target gene expression. NCOR2 is also referred to as a silencing mediator for retinoid or thyroid-hormone receptors (SMRT) or T3 receptor-associating cofactor 1 (TRAC-1).

This gene encodes a nuclear receptor co-repressor that mediates transcriptional silencing of certain target genes. The encoded protein is a member of a family of thyroid hormone- and retinoic acid receptor-associated co-repressors. This protein acts as part of a multisubunit complex which includes histone deacetylases to modify chromatin structure that prevents basal transcriptional activity of target genes. Aberrant expression of this gene is associated with certain cancers. Alternate splicing results in multiple transcript variants encoding different isoforms.[provided by RefSeq, Apr 2011]
