Preparation and Storage

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Natural Cytotoxicity Triggering Receptor 3 (NCR3). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Natural Cytotoxicity Triggering Receptor 3 (NCR3). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Natural Cytotoxicity Triggering Receptor 3 (NCR3), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Natural Cytotoxicity Triggering Receptor 3 (NCR3) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

The protein encoded by this gene is a natural cytotoxicity receptor (NCR) that may aid NK cells in the lysis of tumor cells. The encoded protein interacts with CD3-zeta (CD247), a T-cell receptor. A single nucleotide polymorphism in the 5' untranslated region of this gene has been associated with mild malaria suceptibility. Three transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, May 2010]

NCR3: Cytotoxicity-activating receptor that contributes to the increased efficiency of activated natural killer (NK) cells to mediate tumor cell lysis. Engagement of NCR3 by BAG6 also promotes dendritic cell (DC) maturation, both through killing those DCs that did not properly acquire a mature phenotype, and inducing NK cells to release TNFA and IFNG, which promotes DC maturation. Belongs to the natural cytotoxicity receptor (NCR) family. 6 isoforms of the human protein are produced by alternative splicing.
