Reactivity
Human
Specificity
This assay has high sensitivity and excellent specificity for detection of NPL. No significant cross-reactivity or interference between NPL and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between NPL and all the analogues, therefore, cross reaction may still exist in some cases.

Related Product Information for NPL elisa kit
Intended Uses: This NPL ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human NPL. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: NPL ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-NPL antibody and an NPL-HRP conjugate. The assay sample and buffer are incubated together with NPL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NPL concentration since NPL from samples and NPL-HRP conjugate compete for the anti-NPL antibody binding site. Since the number of sites is limited, as more sites are occupied by NPL from the sample, fewer sites are left to bind NPL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NPL concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the N-acetylneuraminate lyase sub-family of (beta/alpha)(8)-barrel enzymes. N-acetylneuraminate lyases regulate cellular concentrations of N-acetyl-neuraminic acid (sialic acid) by mediating the reversible conversion of sialic acid into N-acetylmannosamine and pyruvate. A pseudogene of this gene is located on the short arm of chromosome 2. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Jan 2011]

Uniprot Description

NPL: Catalyzes the cleavage of N-acetylneuraminic acid (sialic acid) to form pyruvate and N-acetylmannosamine via a Schiff base intermediate. It prevents sialic acids from being recycled and returning to the cell surface. Belongs to the DHDPS family. NanA subfamily. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human NPL npl (Catalog #AAA7240436) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7240436 ELISA Kit recognizes Human NPL. It is sometimes possible for the material contained within the vial of "N acetylneuraminate lyase (NPL), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
