Intended Uses: This sandwich kit is for the accurate quantitative detection of human Myosin Heavy Chain 7, Cardiac Muscle, Beta (also known as MYH7) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human MYH7 antibody. MYH7 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human MYH7 Antibody is added and binds to MYH7 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated MYH7 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human MYH7. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

Muscle myosin is a hexameric protein containing 2 heavy chain subunits, 2 alkali light chain subunits, and 2 regulatory light chain subunits. This gene encodes the beta (or slow) heavy chain subunit of cardiac myosin. It is expressed predominantly in normal human ventricle. It is also expressed in skeletal muscle tissues rich in slow-twitch type I muscle fibers. Changes in the relative abundance of this protein and the alpha (or fast) heavy subunit of cardiac myosin correlate with the contractile velocity of cardiac muscle. Its expression is also altered during thyroid hormone depletion and hemodynamic overloading. Mutations in this gene are associated with familial hypertrophic cardiomyopathy, myosin storage myopathy, dilated cardiomyopathy, and Laing early-onset distal myopathy.
