Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Methionine aminopeptidase 2 (METAP2) ELISA Kit employs a two-site sandwich ELISA to quantitate METAP2 in samples. An antibody specific for METAP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any METAP2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for METAP2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of METAP2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Methionine aminopeptidase 2 encoded by METAP2 is a member of the methionyl aminopeptidase family. Methionine aminopeptidase 2 functions both by protecting the alpha subunit of eukaryotic initiation factor 2 from inhibitory phosphorylation and by removing the amino-terminal methionine residue from nascent proteins. Increased expression of METAP2 is associated with various forms of cancer, and the anti-cancer drugs fumagillin and ovalicin inhibit the protein by irreversibly binding to its active site. Inhibitors of this gene have also been shown to be effective for the treatment of obesity. A pseudogene of METAP2 is located on chromosome 2. Several transcript variants encoding different isoforms have been found for METAP2.

The protein encoded by this gene is a member of the methionyl aminopeptidase family. Uniprot Description

Cotranslationally removes the N-terminal methionine from nascent proteins. The N-terminal methionine is often cleaved when the second residue in the primary sequence is small and uncharged (Met-Ala-, Cys, Gly, Pro, Ser, Thr, or Val). The catalytic activity of human METAP2 toward Met-Val peptides is consistently two orders of magnitude higher than that of METAP1, suggesting that it is responsible for processing proteins containing N-terminal Met-Val and Met-Thr sequences in vivo.
