Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Melanocortin-2 receptor accessory protein (MRAP) ELISA Kit employs a two-site sandwich ELISA to quantitate MRAP in samples. An antibody specific for MRAP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MRAP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MRAP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MRAP bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: MRAP encodes a melanocortin receptor-interacting protein. Melanocortin-2 receptor accessory protein regulates trafficking and function of the melanocortin 2 receptor in the adrenal gland. Melanocortin-2 receptor accessory protein can also modulate signaling of other melanocortin receptors. Mutations in MRAP have been associated with familial glucocorticoid deficiency type 2. Alternatively spliced transcript variants have been described.

NCBI Description

This gene encodes a melanocortin receptor-interacting protein. The encoded protein regulates trafficking and function of the melanocortin 2 receptor in the adrenal gland. The encoded protein can also modulate signaling of other melanocortin receptors. Mutations in this gene have been associated with familial glucocorticoid deficiency type 2. Uniprot Description

MRAP: Required for MC2R expression in certain cell types, suggesting that it is involved in the processing, trafficking or function of MC2R. May be involved in the intracellular trafficking pathways in adipocyte cells. Defects in MRAP are the cause of glucocorticoid deficiency type 2 (GCCD2); also known as familial glucocorticoid deficiency type 2 (FGD2). GCCD2 is an autosomal recessive disorder due to congenital insensitivity or resistance to adrenocorticotropin (ACTH). It is characterized by progressive primary adrenal insufficiency, without mineralocorticoid deficiency. 4 isoforms of the human protein are produced by alternative splicing.
