Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique.Antibody specific for MR has been pre-coated onto a microplate. Standards andsamples are pipetted into the wells and any MR present is bound by theimmobilized antibody. After removing any unbound substances, abiotin-conjugated antibody specific for MR is added to the wells. After washing,avidin conjugated Horseradish Peroxidase (HRP) is added to the wells.Following a wash to remove any unbound avidin-enzyme reagent, a substratesolution is added to the wells and color develops in proportion to the amount ofMR bound in the initial step. The color development is stopped and the intensityof the color is measured.

NCBI Description

The recognition of complex carbohydrate structures on glycoproteins is an important part of several biological processes, including cell-cell recognition, serum glycoprotein turnover, and neutralization of pathogens. The protein encoded by this gene is a type I membrane receptor that mediates the endocytosis of glycoproteins by macrophages. The protein has been shown to bind high-mannose structures on the surface of potentially pathogenic viruses, bacteria, and fungi so that they can be neutralized by phagocytic engulfment.[provided by RefSeq, Apr 2011]

Uniprot Description

Function: Mediates the endocytosis of glycoproteins by macrophages. Binds both sulfated and non-sulfated polysaccharide chains. Acts as phagocytic receptor for bacteria, fungi and other pathogens.

Subcellular location: Endosome membrane; Single-pass type I membrane protein. Cell membrane; Single-pass type I membrane protein Ref.11.

Polymorphism: Genetic variations in MRC1 may influence susceptibility or resistance to leprosy in some populations. Particularly, Gly-396 seems to be a risk factor for leprosy when associated with Ala-399 and Phe-407.

Miscellaneous: CRDs 1-3 have at most very weak affinity for carbohydrate. CRD 4 shows the highest affinity binding and has multispecificity for a variety of monosaccharides. At least 3 CRDs (4, 5, and 7) are required for high affinity binding and endocytosis of multivalent glycoconjugates.
