Reactivity
Human
Specificity
This assay has high sensitivity and excellent specificity for detection of GMPPB. No significant cross-reactivity or interference between GMPPB and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between GMPPB and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
0.1 ng/mL
Preparation and Storage
Store all reagents at 2-8 degree C.

Intended Uses: This GMPPB ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human GMPPB. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: GMPPB ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-GMPPB antibody and an GMPPB-HRP conjugate. The assay sample and buffer are incubated together with GMPPB-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GMPPB concentration since GMPPB from samples and GMPPB-HRP conjugate compete for the anti-GMPPB antibody binding site. Since the number of sites is limited, as more sites are occupied by GMPPB from the sample, fewer sites are left to bind GMPPB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GMPPB concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is thought to encode a GDP-mannose pyrophosphorylase. The encoded protein catalyzes the conversion of mannose-1-phosphate and GTP to GDP-mannose, a reaction involved in the production of N-linked oligosaccharides. Alternatively spliced transcript variants encoding distinct isoforms have been described. [provided by RefSeq, Jan 2009]

Product Notes

The Human GMPPB gmppb (Catalog #AAA7241128) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7241128 ELISA Kit recognizes Human GMPPB. It is sometimes possible for the material contained within the vial of "Mannose 1 phosphate guanyltransferase beta (GMPPB), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
