The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Lysine Specific Demethylase 6B (KDM6B). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Lysine Specific Demethylase 6B (KDM6B). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Lysine Specific Demethylase 6B (KDM6B), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Lysine Specific Demethylase 6B (KDM6B) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

JMJD3: Histone demethylase that specifically demethylates 'Lys- 27' of histone H3, thereby playing a central role in histone code. Demethylates trimethylated and dimethylated H3 'Lys-27'. Plays a central role in regulation of posterior development, by regulating HOX gene expression. Involved in inflammatory response by participating in macrophage differentiation in case of inflammation by regulating gene expression and macrophage differentiation. Interacts with TLE1. Component of the MLL4 complex, at least composed of MLL4, ASH2L, RBBP5, WDR5, and KDM6B. By 12-O-tetradecanoylphorbol-13-acetate (TPA) in myeloid leukemia cells. Belongs to the UTX family. 2 isoforms of the human protein are produced by alternative splicing.
