Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Leucyl-cystinyl aminopeptidase (LNPEP) ELISA Kit employs a two-site sandwich ELISA to quantitate LNPEP in samples. An antibody specific for LNPEP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LNPEP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LNPEP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LNPEP bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: LNPEP encodes a zinc-dependent aminopeptidase that cleaves vasopressin, oxytocin, lys-bradykinin, met-enkephalin, dynorphin A and other peptide hormones. The protein can be secreted in maternal serum, reside in intracellular vesicles with the insulin-responsive glucose transporter GLUT4, or form a type II integral membrane glycoprotein. The protein catalyzes the final step in the conversion of angiotensinogen to angiotensin IV (AT4) and is also a receptor for AT4. Alternative splicing results in multiple transcript variants encoding different isoforms. The deduced 944-amino acid polypeptide contains the HEXXH consensus sequence of zinc metallopeptidases as well as an N-terminal hydrophobic region that resembles a transmembrane domain.

Uniprot Description

IRAP: a leucyl/cystinyl aminopeptidase. A type II membrane protein and secreted. Resides in intracellular vesicles together with GLUT4 and can then translocate to the cell surface in response to insulin and/or oxytocin. Degrades peptide hormones such as oxytocin, vasopressin and angiotensin III, and plays a role in maintaining homeostasis during pregnancy. May be involved in the inactivation of neuronal peptides in the brain. Cleaves Met-enkephalin and dynorphin. Binds angiotensin IV and may be the angiotensin IV receptor in the brain. Three alternatively spliced isoforms have been described.
