Intended Uses: The Insulin ELISA is an enzyme immunoassay for the in vitro measurement of Insulin in serum and plasma (heparin- or EDTA plasma).

Principle of the Assay: The Insulin ELISA kit is a solid phase enzyme-linked immunosorbent assay (ELISA) based on the sandwich principle. The microtiter wells are coated with a monoclonal antibody directed towards a unique antigenic site on the Insulin molecule. An aliquot of unknown containing endogenous Insulin is incubated in the coated well with enzyme conjugate, which is an anti-Insulin antibody conjugated with Biotin. After incubation the unbound conjugate is washed off. During the second incubation step Streptavidin Peroxidase Enzyme Complex binds to the biotin-anti-Insulin antibody. The amount of bound HRP complex is proportional to the concentration of Insulin in the unknown. Having added the substrate solution, the intensity of colour developed is proportional to the concentration of Insulin in the unknown.

NCBI Description

After removal of the precursor signal peptide, proinsulin is post-translationally cleaved into three peptides: the B chain and A chain peptides, which are covalently linked via two disulfide bonds to form insulin, and C-peptide. Binding of insulin to the insulin receptor (INSR) stimulates glucose uptake. A multitude of mutant alleles with phenotypic effects have been identified. There is a read-through gene, INS-IGF2, which overlaps with this gene at the 5' region and with the IGF2 gene at the 3' region. Alternative splicing results in multiple transcript variants.
