Background: Insulin-like growth factor-binding protein 3, also known as IGFBP3, is a member of the insulin-like growth factor-binding protein (IGFBP) family. This protein has an IGFBP domain and a thyroglobulin type-I domain. It forms a ternary complex with insulin-like growth factor acid-labile subunit (IGFALS) and either insulin-like growth factor (IGF) I or II. It plays a role in controlling angiogenesis and cell responses in the human corpus luteum by autocrine/paracrine mechanisms. IGFBP3 protein levels decrease during the progression of prostate cancer from benign prostatic hyperplasia (BPH) to its metastatic form.
NCBI and Uniprot Product Information
NCBI Description
This gene is a member of the insulin-like growth factor binding protein (IGFBP) family and encodes a protein with an IGFBP domain and a thyroglobulin type-I domain. The protein forms a ternary complex with insulin-like growth factor acid-labile subunit (IGFALS) and either insulin-like growth factor (IGF) I or II. In this form, it circulates in the plasma, prolonging the half-life of IGFs and altering their interaction with cell surface receptors. Alternate transcriptional splice variants, encoding different isoforms, have been characterized. [provided by RefSeq, Jul 2008]
Uniprot Description
Function: IGF-binding proteins prolong the half-life of the IGFs and have been shown to either inhibit or stimulate the growth promoting effects of the IGFs on cell culture. They alter the interaction of IGFs with their cell surface receptors. Also exhibits IGF-independent antiproliferative and apoptotic effects mediated by its receptor TMEM219/IGFBP-3R. Ref.16
Subunit structure: Interacts with XLKD1
By similarity. Binds IGF2 more than IGF1. Forms a ternary complex of about 140 to 150 kDa with IGF1 or IGF2 and a 85 kDa glycoprotein (ALS). Interacts with HN. Interacts with TMEM219. Ref.10 Ref.12 Ref.16
Subcellular location: Secreted Ref.16.
Tissue specificity: Expressed by most tissues. Present in plasma.
Developmental stage: IGFBP3 levels are higher during extrauterine life and peak during puberty.
Induction: Up-regulated in the presence of IGF1, insulin and other growth-stimulating factors such as growth hormone, EGF and phorbol esters.
Domain: The thyroglobulin type-1 domain mediates interaction with HN.
Post-translational modification: Phosphorylation sites are present in the extracellular medium.
Sequence similarities: Contains 1 IGFBP N-terminal domain.Contains 1 thyroglobulin type-1 domain.

Product Notes
The Human IGFBP-3 igfbp3 (Catalog #AAA355459) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355459 ELISA Kit recognizes Human IGFBP-3. AAA Biotech's IGFBP-3 can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of IGFBP-3 in human serum, plasma, body fluids, tissue lysates or cell culture supernatants. Researchers should empirically determine the suitability of the IGFBP-3 igfbp3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IGFBP-3, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
