Intended Uses: This sandwich kit is for the accurate quantitative detection of Human Granzymes B (also known as Gzms-B) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human Gzms-B antibody. Gzms-B present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human Gzms-B Antibody is added and binds to Gzms-B in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated Gzms-B antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human Gzms-B. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

This gene encodes a member of the granzyme subfamily of proteins, part of the peptidase S1 family of serine proteases. The encoded preproprotein is secreted by natural killer (NK) cells and cytotoxic T lymphocytes (CTLs) and proteolytically processed to generate the active protease, which induces target cell apoptosis. This protein also processes cytokines and degrades extracellular matrix proteins, and these roles are implicated in chronic inflammation and wound healing. Expression of this gene may be elevated in human patients with cardiac fibrosis. [provided by RefSeq, Sep 2016]

Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution. Cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis.
