Principle of the assay: Gzms-B ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Gzms-B antibody and an Gzms-B-HRP conjugate. The assay sample and buffer are incubated together with Gzms-B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Gzms-B concentration since Gzms-B from samples and Gzms-B-HRP conjugate compete for the anti-Gzms-B antibody binding site. Since the number of sites is limited, as more sites are occupied by Gzms-B from the sample, fewer sites are left to bind Gzms-B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Gzms-B concentration in each sample is interpolated from this standard curve.

NCBI Description

Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein encoded by this gene is crucial for the rapid induction of target cell apoptosis by CTL in cell-mediated immune response. [provided by RefSeq, Jul 2008]

Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution. Cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis. By staphylococcal enterotoxin A (SEA) in peripheral blood leukocytes. Inactivated by the serine protease inhibitor diisopropylfluorophosphate. Belongs to the peptidase S1 family. Granzyme subfamily.
