Principle of the assay: FBXO34 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FBXO34 antibody and an FBXO34-HRP conjugate. The assay sample and buffer are incubated together with FBXO34-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FBXO34 concentration since FBXO34 from samples and FBXO34-HRP conjugate compete for the anti-FBXO34 antibody binding site. Since the number of sites is limited, as more sites are occupied by FBXO34 from the sample, fewer sites are left to bind FBXO34-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FBXO34 concentration in each sample is interpolated from this standard curve.

Members of the F-box protein family, such as FBXO34, are characterized by an approximately 40-amino acid F-box motif. SCF complexes, formed by SKP1 (MIM 601434), cullin (see CUL1; MIM 603134), and F-box proteins, act as protein-ubiquitin ligases. F-box proteins interact with SKP1 through the F box, and they interact with ubiquitination targets through other protein interaction domains (Jin et al., 2004 [PubMed 15520277]).[supplied by OMIM, Mar 2008]
